Rationale
Mechanisms that regulate cytosolic [Ca2+] in airway smooth muscle (ASM) cells are of great interest in developing novel bronchodilators in asthma. However, very little is known about how fluctuations in cytosolic [Ca2+] levels in any one cell influences cytosolic [Ca2+] fluctuations in a another ASM cell in a multicellular ensemble of ASM cells. Also, there is currently no data on how factors such as extracellular matrix (ECM) remodeling influence the frequency of [Ca2+] oscillations in healthy and asthmatic cells.
Methods
Here, we used airway smooth muscle cells from healthy and asthmatic donors to understand the role of the ECM in regulating [Ca2+] oscillations in multicellular ASM ensembles. We analyzed the variation in frequency and correlation in [Ca2+] oscillations for healthy and asthmatic ASM ensembles at E= 300Pa (Healthy) and E= 13kPa (remodeled) ECM stiffness.
