An extreme affinity of Cd II towards the interprotein Rad50 zinc hook domain visualizes how a Zn II-to-Cd II swap can affect the target protein's status and thus illuminate the molecular basis of Cd II-induced genotoxicity. This work showed that Cd II binding to the interprotein Rad50 hook domain induces a two-orders-of-magnitude stabilization effect, which disrupts the overall assembly of the domain and may cause dysfunction of the MRN/X complex.
