Alternative polyadenylation can regulate the length of the 3'-untranslated region (3'-UTR) of mRNAs, thus changing the binding sites available for microRNAs and influencing the expression of mRNAs. Poly (A) sites can be recognized and processed by mammalian cleavage factor I (CFIm), a heterodimer comprised of a 25kDa subunit (CFIm25) containing a Nudix domain and one of two large subunits (CFIm59 or CFIm68). CFIm25 binds directly to RNA and is necessary and sufficient to alter the poly (A) site. Deregulation of CFIm25 has been shown to shorten the 3’-UTR of multiple mRNAs and enhance the level of translation. Here we demonstrate that CFIm25 is down-regulated in the lungs of patients and mice with pulmonary fibrosis, with its expression negatively correlated with levels of fibronectin, a marker for pulmonary fibrosis.
