The term pharmacophore has been used in medicinal chemistry for many years. The official 1998 IUPAC definition1 is as follows:
“A pharmacophore is the ensemble of steric and electronic features that is necessary to ensure the optimal supramolecular interactions with a specific biological target structure and to trigger (or to block) its biological response.” The term is unfortunately also used incorrectly in medicinal chemistry, as implied in the notes that accompany the above definition: 1
