METHODS: ALI was induced in C57/BL6J and IL-10-/-mice via intratracheal (IT) instillation of E. coli lipopolysaccharide (LPS)(O55: B5, 5µg/g). Recombinant IL-10 (50 ng/g) or vehicle was instilled IT 24 hours after LPS administration. Capillary leak was assessed by bronchoalveolar lavage (BAL) measurement of 70 kD FITC-dextran, injected retroorbitally two hours prior to sacrifice. Inflammation was assessed by BAL fluid and whole lung digest cell counts. Lung injury was qualitatively assessed by histopathology.
RESULTS: Peak lung injury occurred at day 4 post LPS instillation. IL-10-/-mice developed more severe lung injury than did wild type mice. IT administration of recombinant IL-10 24 hours after induction of ALI in IL 10-/-mice resulted in decreased capillary leak, decreased inflammatory cell influx, and decreased histopathologic evidence of ALI on day 4 post-LPS treatment.
