Objectives: Pregnancy pathologies, such as preeclampsia (PE) or intrauterine-growth restriction (IUGR) have been linked to placental defect. These diseases can have dramatic consequences on maternal and/or foetal health later in life. The aim of this work is to define loci on the placental genome that genetically regulate changes in the placental transcriptome in a cohort of human samples by mean of an expression Quantitative Trait Loci analysis. These preliminary results highlight regulatory regions of placental function.
Methods: Placental samples (17 control, 6 PE, 3 PE+ IUGR and 10 IUGR) were analysed with ClariomD (Affymetrix) and Infinium OmniExpress (Illumina) arrays in order to obtain both the transcriptome and the genotype of more than 710K SNPs for each sample. 10559 transcripts and 438590 SNPs (MAF> 0.15) were input in MatrixeQTL to perform multivariate linear regression for each gene-SNP …
