Lignin peroxidase (LiP) and manganese peroxidase (MnP) enzymes are the most pervasive type of enzymes that are produced by various types of microorganisms. Bacillus sps strains having the ligninolytic potential was used in this study for the analysis of the LiP and MnP enzyme activity in the presence of various chemical compounds. The major objective of this study is to analyse the activity of the lignin peroxidase (LiP) and manganese peroxidase (MnP) enzyme produced by ligninolytic bacteria Bacillus sps in the presence of different metal salts, solvents and inhibitors and the activity was measured in different ranges of pH. Initially, the enzyme activity of Lip and MnP is 227 U/L and 9.5 U/L respectively. In the presence of various metal ions, the LiPactivity was less affected by the FeSO4 (46.1%) and highly affected by the BaCl2 (77.9%). MnP enzyme activity increased when adding the CuSO4. 5H2O (194.3%) and BaCl2 suppressed the activity by 88.8%. In the inhibitor and metal salts used in the study the L-cysteine and BaCl2 have high enzyme inhibition activity on LiP by 92.6% and 77.9% respectively. In the presence of solvents, the LiP enzyme activity completely suppressed by the ethyl acetate and least inhibit by the chloroform. And in the MnP enzyme activity, ethanol has a high negative effect on the enzyme activity and methanol has less effect. Thus, these findings will be helpful at the time usage of these enzymes in the industries such as biofuel production industries, bioremediation process.
