Psoriasis is chronic inflammatory disease of the skin. In recent years, it has been associated with metabolic diseases, including cardiovascular diseases, diabetes, obesity, and arterial hypertension. Consequently, psoriasis has been proposed as independent risk factor for the metabolic syndrome. However, the common molecular mechanisms driving both diseases in parallel are still largely elusive. 12/15-lipoxygenase (12/15-LO), an enzyme exerting pleiotropic actions, has been suggested to be an important driver of the metabolic syndrome, promoting the pathogenesis of cardio-cerebrovascular disease, diabetes, obesity, arterial hypertension, and non-alcoholic steatohepatitis. The enzyme and its major products, 12 (S)-and 15 (S)-HETE are also elevated in psoriatic lesional skin as well as in the urine of psoriasis patients.
