Epidermolysis bullosa acquisita (EBA) is an autoimmune disease caused by autoantibodies against collagen 7. In the inflammatory form, these autoantibodies fix complement and induce neutrophil influx, subsequently leading to inflammation at the basement membrane zone. To better understand the activation of neutrophils in the blood and how these correspond with lesional neutrophils, we performed single-cell RNA sequencing of blood and skin from passive transfer EBA mice and controls. Dimensionality reduction analysis of blood cells identified a unique population of neutrophils in the EBA group consistent with activated neutrophils. Bioinformatic gating was then performed to isolate neutrophils based onbona fide putative markers, with resolution decreased to consider neutrophils as a single population for each condition.
