Boys with DMD have an increased incidence of neuropsychiatric disorders such as ADHD, anxiety and autism. Interleukin (IL)-6 is raised in DMD patients and mdx mice. We examined mdx behaviour in an open field and determined if neutralisation of central IL-6 signalling normalised mdx behaviours. An intrathecal injection of monoclonal anti-IL-6 receptor antibodies (xIL-6R, 0.2 mg/kg) or saline was administered to wildtype (WT) and mdx mice (8-12 weeks), 24 hours prior to a 10-minute open field behavioural test. Central area frequency (CAF), rearing frequency (RF), faecal output (FO), grooming frequency (GF), freezing frequency (FF) and jumping frequency (JF) were studied. Statistical analysis included unpaired t tests and Welch’s correction when normality was not passed. GF, FF and JJ were similar (n= 19-20, p> 0.05) between saline and xIL-6R groups.
