Abstract
The in vitro screening of 10 potato cultivars (White Desiree, Russet Burbank, Cosima, Agria, Concord, Diamant, Maradona, Kennebec, Marfona and Main) for salt tolerance at different concentrations of NaCl (0, 30, 60, 90 and 120 mM) was conducted. To select the salt-tolerant and salt-sensitive cultivars, shoot length, root length, shoot dry weight (SDW), root dry weight, leaf area, number of rooted shoots (RS) and RDW/SDW were measured. Kennebec and Concord were salt- tolerant and salt-sensitive, respectively. A dendrogram based on dry weight resulted in the clustering of the potato cultivars into 3 groups, i.e. the salt tolerant (Kennebec, Main and Cosima), moderately salt tolerant (White Desiree, Russet Burbank, Maradona and Diamant) and salt- sensitive cultivars (Agraria, Marfona and Concord). RAPD analysis was also conducted using 10-mer random primers. Dendrograms based on DNA banding pattern showed polymorphism by FPK2-19 primer. The banding pattern of RAPD confirmed the distinct polymorphism between salt-sensitive and salt- tolerant cultivars. The clustering pattern of the potato cultivars suggested salt tolerance and salt sensitivity in some potato cultivars are due to genotype variation and not on epigenetic adaptation to salt stress condition.
