A cetyltrimethylammonium bromide-based method to extract low-molecular-weight RNA from polysaccharide-rich plant tissues

In summary, we have adapted the CTAB method to the extraction of LMW RNA from
polysaccharide-rich plant tissues. LMW RNA obtained by the method presented is pure and
intact, and can be loaded in large amount in denaturing PAGE. Thus, it can be used for
siRNA detection and miRNA expression analysis by northern blotting, and to produce small
RNA libraries from plant organs, such as fruits and seeds, in which the role of these small
RNA molecules has been, thus far, little explored.
