We present a resource of nearly two thousand mitochondrial proteins, identified by
combining manual literature curation with machine-learning across multiple experimental
and computational data resources. The resource documents 1,357 genes encoding verified

Abstract
We present a resource of nearly two thousand mitochondrial proteins, identified by combining manual literature curation with machine-learning across multiple experimental and computational data resources. The resource documents 1,357 genes encoding verified mitochondrial proteins (with 1,761 PubMed citations); 127 genes encoding proteins associated with mitochondria (with 111 citations); 485 genes encoding extra-mitochondrial ancillary proteins affecting mitochondrial function (with 540 citations); and 117 genes encoding proteins without publications of mitochondrial localisation or functional impact, but possessing highly significant evidence of mitochondrial localisation from large-scale data sets. Among verified mitochondrial proteins: 9% had multiple subcellular localisations; nearly 50% lacked a mitochondrial targetting sequence; and 39% showed differential tissue expression. GFP-tagging confirmed mitochondrial localisation of several predicted mitochondrial proteins, including SLC30A9/HUEL/C4orf1, which is associated with a cerebro-renal syndrome having hallmarks of a multisystem mitochondrial disease. Our resource aids genetic diagnoses, and provides new opportunities for studying mitochondrial evolution and tissue specificity of mitochondrial proteins and diseases.
